4get (IL-4 green-enhanced transcript)
An IRES-enhanced GFP introduced downstream of the endogenous IL-4 gene. Fluorescent cells are constitutively competent for IL-4 expression – fluorescence increases ~20-fold with activation.

KN2 (Knock-in human CD2 IL-4 reporter)
A non-signaling, nonbinding human CD2 cDNA replaces IL-4 at the ATG start site. Positive cells represent cells that have actively secreted IL-4 as revealed by human CD2 on the membrane. Homozygous mice are IL-4 null.

A loxP-flanked stop cassette followed by diphtheria toxin-alpha introduced into the ubiquitously expressed ROSA26 locus. Expression of Cre recombinase results in excision of the stop cassette and establishes toxin read through and death of the cell.

Floxed IL-4/IL-13
Bacterial recombineering used to engineer a loxP site into the second intron of the IL-13 gene and an FRT/loxP site into the 3’-UTR of the IL-4 gene. Cre expression results in deletion of IL-4 and IL-13 expression through excision of the intervening 21 kB fragment.

ChiaRed mice, useful in studies of chitin-induced inflammation and age-related lung fibrosis, express tdTomato and humanized cre recombinase (hCre; iCre) from the mouse Chia1 (AMCase) gene. Endogenous expression of Chia1 is knocked out in this allele.
